Beta-lactams are the most widely produced and used antibacterial drugs in the world. All of the drugs in this group contain a beta-lactam ring in their structure. A beta-lactam ring is a four-membered lactam. Penicillin derivatives, cephalosporins, monobactams and carbapenems all belong to this popular class of drugs. Mode of Action Beta-lactams exert their bacteriocidal effects in the following ways: *Bind to penicillin-binding proteins (PBPs) β-lactams bind to PBPs because they are similar in chemical structure. * Inhibit synthesis of peptidoglycan Peptidoglycan is the major component of bacterial cell walls. Bacterial cell wall synthesis is essential to growth, cell division (thus reproduction) and maintaining the cellular structure in bacteria. * Excite the release of autolysin An autolysin is an enzyme that hydrolyzes the components of a biological cell. Autolysins exist in all bacteria containing peptidoglycan. *Lyse cells and then kill bacterial cells. Autolysins are naturally produced by peptidoglycan containing bacteria, but excessive amounts will cause the cell to burst due to osmotic pressure.
